公司開發(fā)出IgG全長(zhǎng)抗體酵母展示技術(shù)平臺(tái)，展示的IgG抗體在分子結(jié)構(gòu)、理化特性 和生物學(xué)功能方面與天然抗體相近。這項(xiàng)技術(shù)可以構(gòu)建大容量IgG全長(zhǎng)抗體展示庫(kù)，通過(guò)FACS分選技術(shù)對(duì)抗體親和力及表達(dá)水平同時(shí)進(jìn)行篩選，選出的抗體可以直接進(jìn)行動(dòng)物實(shí)驗(yàn)和臨床試驗(yàn)。在抗體成熟優(yōu)化應(yīng)用中，這項(xiàng)技術(shù)比噬菌體展示更具優(yōu)勢(shì)，噬菌體展示只能展示ScFv等抗體片段，通過(guò)結(jié)構(gòu)轉(zhuǎn)換形成的IgG抗體可能會(huì)失去活性。我們提供涵蓋抗體人源化、親和力成熟、表達(dá)生產(chǎn)在內(nèi)的抗體基因工程改造一站式服務(wù)。

Hangzhou Genekine Biotech Co. Ltd has developed proprietary platforms of glycoengineered pichia pastoris and full-length IgG pichia display system. There are many advantages of these two platforms. Similar to mammalian cell (HEK293) expression system, glycoengineered pichia pastoris contains eukaryotic internal quality control apparatus for protein correct folding and human-like N-glycosylation. Full-length IgG is displayed on pichia surface as close to native form in molecular structure, biophysical property, and biological function.

FACS sorting for IgG pichia display system is based on antibody affinity and display level. The isolated antibody can be directly expressed for animal study and clinical trial. Selection of IgG in pichia display has no need of format conversion. However selection of ScFv in phage display requires format conversion into full-length IgG, which may cause the lose of activity. Unlike the mammalian cell display, our platform can operate in large library size, easy genetic manipulation, fast generation time, and low cost screening. We will provide one-stop shop service for antibody humanization, affinity maturation, and antibody production.
